A novel biotinylated diazirinyl ceramide analogue for photoaffinity labeling OAK

A novel photoreactive ceramide analogue, which contains (3-trifluoromethyl) phenyldiazirinyl lipid and biotinylated sphingosine, was synthesized. The probe was recognized as an antigen by anti-ceramide antibody and as a substrate for sphingolipid ceramide N-deacylase. These results indicate that the probe may be useful as a photoaffinity-biotinylating agent in sphingolipid studies.

A novel neuropeptide Y neuronal pathway linking energy state and reproductive behavior OAK

Animals consume energy for reproduction, as well as survival. Excess or insufficient energy investment into reproduction, respectively, threatens the survival of parents or leads to the failure of reproduction. Management of energy consumption in reproduction is important, not only for the success of the process, but also for the survival of the parents. Reproductive behaviors, such as mating and parental behavior, are indispensable for achieving each event of reproduction including gametogamy, parturition, and lactation. Therefore, reproductive behavior is one of the important factors in managing energy consumption for reproduction. A PCR Based Survey of Babesia ovata in Cattle from Various Asian, African and South American Countries OAK

Babesia ovata is a tick-transmitted hemoprotozoan parasite that infects cattle. In our study, bovine blood samples (n=2,034) were collected from 10 different countries (Brazil, China, Ghana, Japan, Mongolia, the Philippines, South Africa, Sri Lanka, Thailand and Vietnam) and DNA extracted. The DNA samples were screened using an established and specific polymerase chain reaction (PCR) assay targeting the Apical membrane antigen 1 (AMA-1) gene. Parasite DNA was detected among samples collected from Japan, Mongolia and Thailand. Sequence analyses confirmed that the PCR assay detected only B. ovata AMA-1, and that amplicons from different geographical locations were conserved. A PCR-based survey of animal African trypanosomosis and selected piroplasm parasites of cattle and goats in Zambia OAK

We screened cattle and goats from the districts of Chama, Monze and Mumbwa in Zambia for animal African trypanosomes, Babesia bigemina and Theileria parva using PCRs; 38.1% of the samples tested positive for at least one of the parasite species. The most common parasite was Trypanosoma vivax (19.8%). Its incidence was significantly higher in goats than in cattle, (P<0.05). B. bigemina was found in samples from all the three areas, making it the most widespread of the parasites in Zambia. Among the tested samples, 12.0% of the positive samples were mixed infections. Ultrasonography (US) has been applied to the ovary and the uterus of domestic animals from the late 1980s, and established in 1990s as a practical tool for animal production. US made it possible to detect pregnancy at a very early stage and, most importantly, to observe the real-time dynamics of follicular development and hence the discovery of follicular waves. This has greatly contributed to our understanding of ovarian physiology and helped us to develop several "pin-point" protocols for hormonal treatment. A Preliminary Study of 13C-Phenylalanine and 13C-Dipeptide Breath Tests in Horses OAK

This study aimed to establish a standard dose and sample collection time for 13C phenylalanine and 13C-Dipeptide breath test in horses. To evaluate dose-dependent effects, healthy horses received 2.5 mg/kg, 5 mg/kg, and 10 mg/kg 13C phenylalanine dissolved in 1 ml/kg distilled water and 1.25 mg/kg, 2.5 mg/kg, and 5 mg/kg 13C dipeptide dissolved in 2 ml/kg distilled water. Tmax was observed during the sample collection time. For 13C phenylalanine, the standard deviation of Cmax at 5 mg/kg was lower than that of 10 mg/kg. For 13C dipeptide, the standard deviation of Tmax was the lowest at 5 mg/kg. This study revealed that an optimal dose for breath tests with 13C phenylalanine and 13C dipeptide may be 5 mg/kg in horses.

A preliminary study of communal nesting of Siberian flying squirrels Pteromys volans in Japan OAK

To maintain local populations of the Siberian flying squirrel Pteromys volans in restricted small habitats, it is necessary to understand their nesting and food biology. Communal nesting of P. volans is well documented in the Eurasian continent, but the details of this behavior have been poorly investigated in Japan. We investigated a group composition of P. volans in Hokkaido, Japan. The mean numbers of individuals using each nest box and tree cavity were 2.17 and 2.44, respectively. Although each female had its own exclusive home range, communal nesting composed by two females was observed in nest boxes. A preliminary study on origin of Callosciurus squirrels introduced into Japan OAK

To examine the origin of Callosciurus erythraeus introduced to Japan, we compared mitochondrial DNA control region sequences of eight haplotypes from Japan with those of 42 haplotypes from Taiwan. There were two distinct phylogroups in Japan. Six haplotypes from Japan were included in the Taiwan population, suggesting that they could have been introduced from Taiwan. Two haplotypes, however, were distantly related to the cluster consisting of all Taiwanese haplotypes and six Japanese haplotypes. A questionnaire survey of educational programs at the Obihiro Zoo, using Pteromys volans orii—an animal native to Hokkaido—as a subject OAK

日本の動物園の二大事業として「種の保存」および「環境教育」が掲げられている.今後の動物園における教育的取り組みのさらなる充実が期待され,アンケート調査などを用いた来園者の実態調査および他機関との連携が不可欠とされている.おびひろ動物園で飼育されているエゾモモンガPteromys volans orii は,市街地の緑地などに生息する比較的身近な動物であるものの,夜行性の小型動物であるため一般市民の認知度は低い.一方,帯広畜産大学ではエゾモモンガに関する様々な研究がなされており,多くの知見が積み重ねられている.また本種は外見から人々に好印象を与える特徴を持っており,教育の題材として受け入れられやすいと考えられる.そこで本研究では,おびひろ動物園の来園者に動物園に対する意識およびエゾモモンガについてのアン ケート調査を行ない来園者の実態を知ることで,より効果的な教育への提言を行なうことを目的とした.調査の結果,年齢や性別,来園時のグループ構成および来園目的の違いは「エゾモモンガを見たことがあるか」,「エゾモモンガが身近に住む動物であることを知っていたか」に関係し ていなかったものの,自然保護活動に対する興味が強い人ほどこのような経験や認識が豊富であることが明らかになった.また,帯広畜産大学とおびひろ動物園の連携について, 95.4%の来園者が重要であると解答していた.したがって,郷土の動物であるエゾモモンガを用いて大学と動 物園が協力して教育を行なっていくことで,帯広市民の自然に対する興味関心を増大させることができる可能性があると考えられる.

A Review of Neospora caninum and Neospora-like Infections in Animals OAK

Neospora caninum is a recently recognized protozoan parasite of animals. Until 1988, it was misdiagnosed as Toxoplasma gondii. Neospora caninum or Neospora-like parasites cause paralysis and death in dogs and neonatal mortality and abortion in cattle, sheep, goats and horses. Its life cycle is not known. Tachyzoites and tissue cysts are the only asexual stages known and the carnivorous definitive host is not known. Transplacental transmission is the only known natural route of infection.

A Review of the Taxonomy of Theileria sergenti/buffeli/orientalis Group Parasites in Cattle OAK

The controversial circumstances relating to the classification of the benign Theileria species from Japan, Australia and Britain, which are frequently referred to as T. sergenti/buffeli/orientalis group parasites, was reviewed. Our recent systematic comparisons suggest a possibility that the Japanese T. sergenti (minor piroplasm in common parlance) might be a new species and the Australian T. buffeli and British T. orientalis might belong to one and the same species. The necessity for the review of the genus name for these benign Theileria species was also briefly discussed after the morphological studies of their schizogony.
